3 seconds is enough.
The 3-500z tubes are basically "instant-on".
However, I do not turn the amp off immediately.
If I have used it long enough for the cabinet of the amp to be
hot, I let it idle for a long time, giving the tubes a chance to
cool off a bit before I turn the amp off. If you turn it off
immediately, the fan stops blowing (of course) and the tubes can
stay very hot for a long time.
I've been running 3-500z tubes for over 30 years and have never
had any problems, even though I begin using them immediately.
