Connect the hardware PTT (VOX) from SignaLink to the PKS pin (pin 9)
of the ACC2 connector and disable COMM PTT (software command PTT) in
RTTY.
The TS-2000 (and other recent Kenwood transceivers) requires PTT on
the PKS pin to enable audio from the ACC2 jack (and mute the mic)
for AFSK operation. The software PTT commands emulate PTT on the
mic jack and enable the microphone.
It's also possible that Windows 7 has the audio output level turned
down for WL/MMTTY but the first thing to check is proper PTT to
select the ACC2 input.
